Affron® Saffron Extract: The Clinical Science Behind Mood Mod

Affron is the standardized saffron extract used in most published human trials on saffron for mood. Here is what it is, the dose the research is built on, how it works, and the studies behind it, alongside the rest of the Mood Mod formula.

What is Affron® saffron extract?

Affron is a patented saffron extract standardized to consistent levels of its two active compounds, crocins and safranal, the bioactives tied to saffron's mood-supporting effects in research.

Not all saffron is the same. Unlike generic saffron powder, whose active content varies batch to batch, Affron guarantees a consistent level of its actives in every batch, which is why it has been used across multiple peer-reviewed human clinical trials. It is manufactured by Pharmactive Biotech. When you see Affron on a label, you are getting the same extract used in the published research rather than an unstandardized powder. We cover this in more depth in our guide to what Affron is and why it matters.

How It Works

Crocins support the reuptake of dopamine and norepinephrine, while safranal primarily influences serotonin activity. Together, these mechanisms support a more balanced emotional state. We go deeper on saffron for stress and cortisol and saffron for anxiety in our dedicated guides. Because saffron acts on serotonin pathways, anyone on an SSRI or other serotonergic medication should speak with their doctor before use.

Landmark Clinical Trial (2025)

The largest Affron trial to date, published in The Journal of Nutrition in 2025, enrolled 202 adults with subclinical depressive symptoms. Participants taking 28mg of Affron daily for 12 weeks showed significantly greater improvement in mood compared to the placebo group, and reported reduced daily feelings of sadness and stress as early as five weeks into the study. What is the clinical dose of saffron for mood?

The dose used in most positive clinical trials is 28 to 30mg per day of a standardized saffron extract.

Earlier trials have shown that Affron at 28mg per day improved mood and reduced anxiety symptoms, and that an evening dose supported better sleep quality in healthy adults over 4 to 8 week periods. That dose is nearly impossible to reach through cooking, which is why supplementation is the practical route. Mood Mod is formulated with 30mg of Affron per serving, a deliberate margin above the 28mg clinical dose, so every stick pack delivers at least the full studied amount even after accounting for natural manufacturing variation. For a full breakdown, see our guide to the right saffron dosage.

How long does saffron take to work?

Most clinical trial participants noticed meaningful changes within four to eight weeks of consistent daily use, though some reported a shift within the first couple of weeks.

Saffron builds up in the body over time rather than acting immediately, so daily consistency matters more than timing. The L-Theanine and magnesium in the formula can contribute a same-day sense of calm, while the saffron works over the longer window. If you are specifically looking at saffron for mood, the research shows the benefit builds over four to eight weeks. We break down what to expect week by week in our saffron timeline guide.

L-Theanine: calm focus without sedation

L-Theanine is an amino acid from green tea that promotes alpha brain wave activity, the state of feeling calm but mentally sharp, without drowsiness.

Dosage matters. Many supplements use 50 to 100mg of L-Theanine. Mood Mod uses 200mg, which is at the higher end of the effective range studied for stress and cognition. At this dose, L-Theanine pairs well with saffron to create a calm, non-drowsy effect you can rely on any time of day.

How It Works

L-Theanine increases alpha brain wave production, associated with wakeful relaxation. It also supports healthy levels of GABA, serotonin, and dopamine, the neurotransmitters that regulate mood, focus, and stress response. This is why saffron and L-theanine work so well together for focus.

Saffron, L-Theanine & Cortisol

How Mood Mod works on the cortisol stress response.

One of Mood Mod's actives has direct human data on the acute cortisol stress response, at the same 200mg dose in every stick.

In a randomised, double-blind, placebo-controlled crossover trial, a drink containing 200mg of L-theanine reduced the salivary cortisol response to a multitasking stressor, measured three hours after the dose, with a lower subjective stress response at one hour. Two honest caveats: that drink also carried small amounts of three other ingredients, and a separate four-week trial of L-theanine alone found no change in resting cortisol. So this is an acute stress-response effect, not a claim that Mood Mod lowers cortisol generally.

Saffron has its own cortisol evidence, and it is more direct. In a 2026 randomised, double-blind, placebo-controlled trial, a standardised saffron extract significantly reduced serum cortisol at both day 28 and day 56 against placebo. Three things to keep in view: that trial used a different standardised extract, not Affron®; it ran at 15mg, half the 30mg in a Mood Mod stick; and it enrolled 56 adults, so it is a small study. It supports saffron acting on cortisol. It is not, on its own, proof that this formula does.

Magnesium glycinate: the nervous system reset

Magnesium glycinate is a well-absorbed, gentle form of magnesium bound to the amino acid glycine, chosen because glycine itself has calming properties and the chelated form is easier on the stomach.

Over half of Americans do not get enough magnesium from their diet, and stress, caffeine, alcohol, and processed food deplete it further. This matters for mood because magnesium plays a direct role in nervous system function, helping calm NMDA receptors and support GABA. Magnesium also plays a role in saffron for irritability and mood swings. Glycinate is the form most commonly recommended when the goal is supporting calm and sleep quality.

Why Glycinate?

Cheaper forms like magnesium oxide have far lower bioavailability and a stronger laxative effect. Glycinate is bound to glycine, which improves absorption and is gentler on the stomach, making it the preferred form for calm and sleep support.

2.5mg B6 · 25mcg B12

Vitamin B6 + B12: the mood cofactors

B6 and B12 are essential cofactors your body needs to produce serotonin and dopamine, the neurotransmitters that regulate mood, motivation, and emotional stability.

B6 and B12 are essential cofactors in the production of serotonin and dopamine. Many people run low on these vitamins, especially those under chronic stress, consuming alcohol regularly, or taking certain medications, so Mood Mod includes both to support a more stable mood baseline.

How They Work

B6 is required for the enzymatic conversion of 5-HTP into serotonin. B12 supports dopamine synthesis and energy metabolism. Both are water-soluble, so your body does not store them and needs a consistent daily supply.

Electrolytes: hydration meets mood

Even mild dehydration can affect mood, energy, and focus, so Mood Mod includes an electrolyte blend of sodium and potassium to support daily hydration at the cellular level.

Supporting hydration supports the biological systems responsible for emotional regulation. It also makes Mood Mod something you actually look forward to drinking every day, which matters for consistency, and consistency is what drives results with this formula.
